Prof Martin Herrmann, University Clinic Erlangen, Friedrich-Alexander University of Erlangen-Nürnberg discusses the role of NETs in human disease.

NETs are decondensed chromatin filaments ejected by the nuclei of neutrophil granulocytes upon stimulation. NET release is an evolutionary stable function of the innate immune system of all mammalians. NET formation must generally be considered beneficial to the host.

However, NETs propel pathological progression of a plethora of diseases. DNA drives the interferon system and forms the scaffold for proteins of the innate immune system. An insufficient clearance of extruded chromatin can perform a secret task of moonlighting in occlusive disorders.

I will discuss (1) events involved in chromatin release (2) devastating consequence of a dysregulated NET formation, and (3) the imbalance between NET formation and clearance. I focus on the role of NETs in the occlusion of vessels and ducts. To develop new therapies, it is important to target chromatin decondensation during NET formation and aggregation. Therapies that support the clearance of extracellular chromatin are also conceivable.
